The Saints went out of the historic cup at this stage last season, losing to fellow north Londoners Tottenham.
Arsenal, meanwhile, won the cup and have been in good form as of late.
The winner of this tie will face Wolves in the fifth round following their win over Chorley.

Southampton v Arsenal: Team news
Southampton striker Danny Ings is back in contention for the visit of Arsenal.
Ings, who recently recovered from a hamstring injury, has been in isolation after testing positive for coronavirus and missed the Saints’ last two matches.
Southampton made seven changes for the midweek cup win against Shrewsbury and still have a lengthy injury list, which includes latest casualty Will Smallbone, who will be out for six months after damaging knee ligaments.
Arsenal, meanwhile, could hand a debut to Mat Ryan after the goalkeeper joined on loan from Brighton on Friday.
The Gunners are likely to shuffle their pack and Ryan could come straight in for Bernd Leno.
Pablo Mari misses out but Dani Ceballos will undergo a late fitness test on his own calf complaint and could be involved.
Southampton v Arsenal: What has been said?
Southampton boss Ralph Hasenhuttl has challenged his side to beat Arsenal twice in four days as they prepare to meet in the cup first this weekend and then league on Tuesday.
“I’d prefer winning both to be honest, if it’s possible we’ll do it, but I know it’s tough,” the 53-year-old Austrian said.
“Yes, we have the chance to I think. On our day we can beat any team in the league, we’ve shown this, especially at home.
“I think this is the goal we have. Yes, tough like always, but not impossible.”
If Southampton beat Arsenal on Saturday and progress to the fifth round, they will play six matches in 19 days, but Hasenhuttl is not about to sacrifice the cup.
“It’s a busy time now with a lot of games coming up. We have an FA Cup week, if you want, two games in a week in the cup,” he said.
“It’s a new challenge and in this competition we want to go as far as possible.”
